Does it matter if the transmission (545-rfe) came out of a 2wd or 4wd? Do I need one with a certain spline count?

They're all the same, it doesn't matter because you'll be replacing either the 4wd adapter or the 2wd tail cone with the 4wd adapter from the 3/4 ton truck. Only difference would be if you can get your hands on a 3/4 ton trans, you could skip that step and the cost of the adapter.
